Retatrutide (CAS 2381089-83-2) is a new-generation synthetic peptide analogue, described in the clinical and pharmacological literature as a triple agonist — a single molecule that simultaneously activates three receptors: GLP-1 (glucagon-like peptide-1), GIP (glucose-dependent insulinotropic polypeptide) and GCGR (the glucagon receptor). This triple action sets retatrutide apart from incretin peptides that act on one or two receptors and is the basis of the scientific interest in the molecule in the fields of metabolism and GLP-1, GIP and glucagon receptor signalling.
Unlike the single agonist Semaglutide (GLP-1 only) and the dual agonist Tirzepatide (GLP-1 + GIP), retatrutide adds glucagon activity. It is precisely the glucagon component that is linked in the published literature with activation of the glucagon receptor (GCGR) — beyond activation of the GLP-1 and GIP receptors.
Retatrutide is being studied in several therapeutic areas: obesity (Phase II, published in the New England Journal of Medicine, 2023), type 2 diabetes (Phase II, The Lancet, 2023), metabolic dysfunction-associated steatotic liver disease (MASLD/NASH, Phase IIa, Nature Medicine, 2024), as well as in the ongoing, large-scale Phase III TRIUMPH programme. At present retatrutide is an experimental drug candidate — it is not approved by the FDA or the EMA for clinical use. See also the other peptides for weight loss — GLP-1 agonists in our category.
In the Peptology version, retatrutide is supplied as a lyophilised research peptide with purity above 99%, confirmed by independent laboratory analysis (CoA) for each batch. It is available in four vial sizes — 5 mg, 10 mg, 15 mg and 30 mg, depending on the scale of the research protocol. It is NOT an approved medicinal product — the product is intended exclusively for scientific, research and in vitro applications.
In clinical and preclinical publications, retatrutide is described as a molecule with three simultaneous receptor actions. These three mechanisms work in a coordinated way and are described in the literature as simultaneous signalling at the GLP-1, GIP and glucagon receptors — a combination that is not achieved with incretin peptides that act on one or two receptors.
GLP-1
Glucagon-like peptide-1 — in the literature it is linked with activation of the GLP-1 receptor in central pathways, slowing of gastric emptying and glucose-dependent insulin secretion.
GIP
Glucose-dependent insulinotropic polypeptide — studied in relation to improved insulin sensitivity and an additional metabolic action that complements the GLP-1 component.
GLUCAGON (GCGR)
Glucagon receptor — in the published data, activation of the glucagon receptor (GCGR) and intracellular cAMP signalling are described; this is the component that sets retatrutide apart.
Most incretin peptides act only on the GLP-1 receptor (such as Semaglutide) or on the GLP-1 and GIP receptors (such as Tirzepatide). The addition of controlled glucagon activity is a fundamentally different approach — in the literature, glucagon receptor agonist action is described at the level of the receptor (GCGR) and intracellular signalling (cAMP). This makes retatrutide a subject of particular scientific interest in models of metabolic dysfunction-associated steatotic liver disease (MASLD/NASH).
This idea was explored in a Phase IIa study in MASLD, published in Nature Medicine (2024), in which retatrutide was associated with a significant reduction in liver fat content compared with placebo. These results come from controlled clinical trials and are not a guarantee of an individual result, nor a claim about the action of a research-grade product.
Retatrutide is a peptide with a relatively long half-life; published pharmacokinetic studies describe it as a molecule with predictable, linear kinetics.
The table below compares retatrutide with the two most frequently studied incretin peptides in the catalogue. All three molecules are studied for metabolic regulation and body weight control, but they act on a different number of receptors — Semaglutide on one (GLP-1), Tirzepatide on two (GLP-1 + GIP) and retatrutide on three (GLP-1 + GIP + glucagon). The data are presented as scientific reference and do not constitute a direct head-to-head comparison.

Sources: Jastreboff et al., NEJM (2023) for retatrutide in obesity; Rosenstock et al., The Lancet (2023) in type 2 diabetes; Sanyal et al., Nature Medicine (2024) in MASLD; the SURMOUNT (Tirzepatide) and STEP (Semaglutide) programmes. The data presented come from different clinical programmes and should not be interpreted as a direct comparison, nor as a claim about the result of a specific research-grade product.
The clinical programme of retatrutide is developing in several parallel directions — obesity, type 2 diabetes, metabolic dysfunction-associated steatotic liver disease (MASLD/NASH) and the ongoing, large-scale Phase III TRIUMPH programme. The Phase II results are among the strongest reported in the incretin category, but the molecule has not yet received regulatory approval.
In early clinical studies in healthy volunteers and in patients with type 2 diabetes, predictable, dose-proportional pharmacokinetics were documented, together with a long half-life (~6 days), compatible with weekly administration, and an adverse-event profile dominated by dose-dependent gastrointestinal effects — characteristic of the incretin class.
The study, published in the New England Journal of Medicine (Jastreboff et al., 2023), included 338 adults with obesity without type 2 diabetes, who were randomised to retatrutide 1, 4, 8 or 12 mg SC weekly or to placebo for 48 weeks. At the highest dose (12 mg), the mean reduction in body weight reached ~24.2%, while the threshold of a reduction of ≥ 5% was achieved by all participants at 8 mg and 12 mg. Improvements were also observed in cardiometabolic markers — waist circumference, blood pressure, HbA1c, fasting glucose, insulin and lipid profile. These results come from a controlled clinical trial and are not a recommendation for individual use.
In a randomised Phase II study in patients with type 2 diabetes (Rosenstock et al., 2023), retatrutide was associated with dose-dependent reductions in HbA1c and body weight compared with placebo and with an active comparator. The data supported the dual metabolic hypothesis — simultaneous glycaemic control and weight reduction.
In a Phase IIa study in patients with metabolic dysfunction-associated steatotic liver disease, retatrutide was associated with a significant reduction in liver fat content, measured by MRI-PDFF, in a high proportion of participants. The glucagon component is cited as a possible driver of this effect. These are early data, still to be confirmed in later phases.
The large-scale Phase III TRIUMPH programme is evaluating retatrutide in obesity, in type 2 diabetes and in related metabolic conditions, including studies with long-term and cardiovascular endpoints. Until this programme is completed and regulatory evaluation takes place, retatrutide remains experimental and is not approved for clinical use.

Research warning: Retatrutide is offered exclusively for scientific, research and in vitro applications. It is NOT an approved medicinal product and is not intended for the diagnosis, treatment or prevention of disease in humans or animals. The molecule is not approved by the FDA or the EMA, and clinical efficacy and safety in humans have not been documented. All the information on this page is provided as scientific reference and does not constitute medical advice, a recommendation for use or a claim of results. Use is the sole responsibility of the qualified researcher.
The main difference is the number of receptors. Semaglutide acts only on the GLP-1 receptor; Tirzepatide — on the GLP-1 and GIP receptors; while retatrutide adds a third — the glucagon receptor. In the literature, the glucagon component refers to the additional activation of the glucagon receptor (GCGR), which makes retatrutide a subject of distinct scientific interest.
Combinations between metabolic peptides or with cellular energy molecules (e.g. SS-31 or NAD+ ) are a subject of scientific interest because of complementary mechanisms, but have not been systematically studied under controlled clinical conditions. They remain a subject of research and not a recommendation for use.

Retatrutide is available in: 5/10/15/30 mg.
Retatrutide is available in four lyophilised forms, covering different scales of research work — from individual in vitro series and validation experiments to longer protocols with higher peptide consumption.
5 MG
Starter form
Suitable for validation experiments, individual in vitro series and short-term laboratory protocols.
10 MG
Typical form
A balanced choice for repeated cell series and medium-length protocols.
15 MG
Extended form
Suitable for longer research series and protocols with a higher working-solution concentration.
30 MG
Maximum form
Economical for extended protocols, multi-series models and work with a larger volume of material.
All four forms — 5 mg, 10 mg, 15 mg and 30 mg — have the same purity > 99% and an identical laboratory certificate for the corresponding batch. The choice of size depends on the scale of the protocol and not on the concentration of the active substance.
Important: The information below describes the doses used in the published clinical studies of retatrutide. It is provided as scientific reference and is not a recommendation for individual use, medical advice or administration instructions. The research-grade retatrutide offered by Peptology is not an approved medicinal product.
Unlike peptides with a short half-life, retatrutide has a long half-life (~6 days) and in the clinical programmes is administered once a week, with gradual escalation of the dose to limit gastrointestinal effects. The typical titration scheme in the studies follows steps of 4 weeks:
Similar to other GLP-1 peptides, such as Semaglutide and Tirzepatide, retatrutide requires slow titration for good tolerability — in the clinical studies the dose is increased gradually rather than starting directly at the target level. These regimens are part of controlled clinical trials and are not a recommendation for self-administration.
Retatrutide is supplied as a lyophilised (freeze-dried) powder in a vial. In lyophilised form the peptide is stable both in the refrigerator and at room temperature, protected from light and moisture. After reconstitution, stability depends on the solvent used, with Peptide Matrix Water™ significantly extending the shelf life of the peptide.
Tip for laboratory work: When ordering, you can add Peptide Matrix Water™ to the basket — a stabilising solution designed for work with reconstituted research peptides.

BEFORE RECONSTITUTION
Room temperature, protected from direct light, in the original packaging. Refrigeration is not mandatory.
AFTER RECONSTITUTION
+2 °C to +8 °C, up to 60 days with Peptide Matrix Water. Discard the remainder after this period.
FREEZING
Not recommended. Freeze/thaw cycles degrade the structure of the peptide.
1. Shaking for “faster dissolution”
Foam = denaturation. Only gentle swirling between the palms.
2. Unsuitable solvent
For the stability of the peptide structure, use only a sterile, purpose-designed solvent — Peptide Matrix Water is recommended.
3. Ignoring slight cloudiness
If it is not completely clear = the sample does not meet the criteria for work.
4. Removing it from the refrigerator “for an hour or two”
Short exposures add up and jeopardise stability.
Research status: All Peptology products are intended exclusively for laboratory use and research. They are not pharmaceutical products, have not been approved for human or veterinary use and do not replace medical advice. The protocol described is standard laboratory practice for working with lyophilised peptides.
The vial with the lyophilised peptide is stored protected from light and moisture; for long-term storage a refrigerator or freezer is recommended. After reconstitution, stability depends on the solvent: with Peptide Matrix Water™ — up to 60 days in the refrigerator at 2–8 °C; with bacteriostatic or sterile water — up to 28 days. In every case, avoid re-freezing.
99%+ purity, HPLC/MS certificate for every batch.

Retatrutide activates three receptors simultaneously. In the literature, the GLP-1 component is linked with activation of the GLP-1 receptor in central pathways and glucose-dependent insulin secretion; the GIP component — with improved insulin sensitivity; and the glucagon component (GCGR) — with activation of the glucagon receptor (GCGR). The combination of these three pathways differs fundamentally from incretin peptides that act on one or two receptors. These results come from controlled laboratory and clinical studies.
