What is Cagrilintide?

Cagrilintide (AM833) is a synthetic, long-acting amylin analogue — an agonist of the amylin receptors (AMY1, AMY3) and of the calcitonin receptor. The attached fatty acid residue ensures reversible binding to albumin and a long half-life (~7–8 days).

Unlike incretin agonists such as Semaglutide (GLP-1), Tirzepatide (GLP-1/GIP) or Retatrutide, Cagrilintide acts on a separate pathway — the amylin receptors in the CNS. That is why it is studied on its own and in combination with GLP-1 receptor agonists, such as semaglutide. It remains an experimental molecule; you will find the ready-made combination as CS10.

At Peptology it is supplied as a lyophilised research peptide with purity above 99% (HPLC/MS, CoA per batch), in a 5 mg pack. Exclusively for research and in vitro applications; it is not a medicine. See also the peptides for weight loss (GLP-1).

Available pack sizes: 5 mg and 10 mg

Cagrilintide is available in lyophilised 5 mg and 10 mg forms, covering the typical scales of research work — from individual in vitro runs to longer protocols, including schedules in combination with GLP-1 agonists.

5 MG

Standard pack

A lyophilised amylin analogue with purity > 99%, for studies of the amylin receptors (AMY1, AMY3) and calcitonin, on its own or together with GLP-1 agonists.

10 MG

Extended pack

Double the quantity in one vial — for longer series, higher target concentrations and fewer reconstitution cycles.

COMBINATION

CS10 (Cagrilintide + Semaglutide)

A ready-made research combination of amylin + GLP-1 in one vial — for protocols that study the additive effect of the two pathways.

The 5 mg and 10 mg packs have purity > 99% and a laboratory certificate for the corresponding batch. The choice of quantity depends on the scale of the protocol, not on the concentration of the active substance.

How it acts — agonism of the amylin receptors

Cagrilintide mimics the action of amylin by stimulating the amylin receptors in the brain — a pathway separate from the incretins (GLP-1/GIP). The three axes below summarise the main directions.

AMYLIN RECEPTORS

It activates the amylin receptors (AMY1, AMY3) — complexes of the calcitonin receptor with the proteins RAMP1/RAMP3.

SATIETY & APPETITE

It also binds to the calcitonin receptor (CTR); in research models gastric emptying is recorded as a parameter.

SYNERGY WITH GLP-1

Because the amylin pathway is separate from the GLP-1 receptor, it is also studied in combination with semaglutide.

Amylin and GLP-1 act on different receptor families (AMY/CTR and GLP-1R); that is why Cagrilintide is mainly studied together with GLP-1 agonists. The results of the studies are listed in the “Clinical studies” section.

Pharmacokinetics

  • Half-life: ~7–8 days (~180 hours) — once-weekly dosing
  • Long-acting mechanism: reversible binding to albumin (like semaglutide)
  • CAS number: 1415456-99-3
  • Class: amylin analogue, separate from the GLP-1/GIP incretins
  • Phase II doses (monotherapy): 0.3–4.5 mg SC weekly

Cagrilintide vs Semaglutide vs CS10

Cagrilintide is an amylin analogue (AMY/CTR receptors), not an incretin agonist. Scientific comparison data, not a direct head-to-head comparison.

Clinical studies

In phase II (The Lancet, 2021; 706 adults, 26 weeks), cagrilintide as monotherapy (0.3–4.5 mg SC weekly) gave a mean weight reduction of 6.0–10.8% versus 3.0% with placebo. The half-life in early studies was ~180 hours.

In the phase III REDEFINE 1 (NEJM, 2025; ~3400 adults with obesity, 68 weeks), the combination of cagrilintide + semaglutide gave a mean weight reduction of 20.4%, versus 11.5% for cagrilintide alone, 14.9% for semaglutide alone and 3% for placebo; ~60% of participants achieved a reduction of ≥ 20%. The parallel REDEFINE 2 evaluated the combination in type 2 diabetes, with a significant weight reduction and improvement in glycaemic control.

Cagrilintide is available in: 5/10 mg.

Research protocols

Important: The doses relate to published clinical studies, for scientific and informative purposes — not a recommendation for use or medical advice. Research-grade Cagrilintide is not an approved medicine.

In the clinical programmes, cagrilintide was used weekly, subcutaneously, with gradual titration (phase II: 0.3–4.5 mg SC, titration up to 6 weeks; phase III REDEFINE: cagrilintide + semaglutide). Like the GLP-1 agonists (Semaglutide, Tirzepatide), it requires slow titration; you will find the ready-made combination as CS10. Part of controlled studies, not a recommendation for stand-alone use.

Storage and reconstitution

  • Before reconstitution: room temperature, protected from light and moisture; refrigeration is not mandatory.
  • With Peptide Matrix Water™: in the refrigerator at +2 to +8 °C for up to 60 days.
  • With bacteriostatic/sterile water: in the refrigerator at +2 to +8 °C for up to 28 days; do not re-freeze.
  • Reconstitution: add the solvent slowly down the wall (max. 3 mL); swirl gently, do not shake.

Tip: add Peptide Matrix Water™ — a stabilising solution for reconstituted research peptides.

Laboratory protocol — reconstitution, research protocols, storage

Standard protocol, applicable to all research peptides in the Peptology catalogue. For the concentration of the solution (mg/mL) per vial, use the peptide reconstitution calculator.

The 60-second protocol

2. RECONSTITUTION

The solvent is added to the wall of the vial, without agitation, then wait 10–15 minutes.

3. CALCULATION

Calculator → concentration in mg/mL

4. STORAGE

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water

Quick reference

Parameter Value
Form Lyophilised powder
Solvent Peptide Matrix Water
Volume for reconstitution 1–3 mL — 3 mL maximum (capacity of the standard vial)
Storage before reconstitution Room temperature, protected from direct light (refrigeration is not mandatory)
After reconstitution Up to 60 days at +2 °C to +8 °C with Peptide Matrix Water

What you will need

Precision pipette

Calibrated, 100–1000 µL — for accurate measurement of the volume of solvent added to the vial.

70% alcohol pads

Two pads — to disinfect the peptide vial and the solvent vial.

Peptide Matrix Water

Stabilised solvent for lyophilised peptides. Why this one.

Five rules for reconstitution

✅ Correct ❌ Wrong
Disinfect the stoppers and the work surface with 70% alcohol, work with gloves Working without disinfection
The solvent is added slowly down the wall of the vial Direct jet onto the powder — foaming and denaturation
Light swirling between the palms Shaking — foam, on which the peptides denature
Wait 10–15 minutes at room temperature Mechanical mixing or heating
Visual inspection: clear, without particles Using a cloudy or coloured solution

Calculating the concentration

⚠️ Maximum 3 mL of solvent. The standard vial does not allow a larger volume — do not exceed it during reconstitution.

  • Concentration (mg/mL) = mg in the vial ÷ mL of solvent
  • Volume per sample (mL) = amount per sample (mg) ÷ concentration (mg/mL)
  • 1 mg/mL = 1 µg/µL · 1 mL = 1000 µL

Reconstitution per vial — solvent: Peptide Matrix Water. Concentration after addition (1 mg/mL = 1 µg/µL):

Vial+ 1 mL+ 2 mL+ 3 mL
5 mg5 mg/mL2.5 mg/mL1.67 mg/mL
10 mg10 mg/mL5 mg/mL3.33 mg/mL

👉 For any vial and solvent volume — use the calculator. It calculates the concentration (mg/mL, µg/µL) and the volume per sample (mL, µL).

Storage

BEFORE RECONSTITUTION

Room temperature, protected from direct light, in the original packaging. Refrigeration is not mandatory.

AFTER RECONSTITUTION

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water. Discard the remainder after this period.

FREEZING

Not recommended. Freeze/thaw cycles degrade the structure of the peptide.

Four mistakes to avoid

1. Shaking for “faster dissolution”

Foam = denaturation. Only gentle swirling between the palms.

2. Unsuitable solvent

For the stability of the peptide structure, use only a sterile, purpose-designed solvent — Peptide Matrix Water is recommended.

3. Ignoring slight cloudiness

If it is not completely clear = the sample does not meet the criteria for work.

4. Removing it from the refrigerator “for an hour or two”

Short exposures add up and jeopardise stability.
