What is Eloralintide?
Eloralintide 10 mg — selective amylin receptor agonist
Eloralintide (CAS 2883634-40-8) is a research peptide from the new generation of amylin analogues. Unlike most molecules in the peptides for weight loss category, it does not act through the GLP-1 receptor, but is designed as a selective amylin receptor agonist — the hormonal system that signals to the brain that the meal is over.
Amylin is secreted together with insulin by the pancreatic β-cells. However, the natural hormone breaks down within a few minutes, and its first analogues required several injections a day. Eloralintide was developed as a long-acting molecule for once-weekly administration, with a preference for the amylin receptor over the calcitonin receptor.

Mechanism: amylin receptor signalling without GLP-1
Several parallel hormonal signalling systems are described in the literature. Incretin peptides (semaglutide, tirzepatide) act through the GLP-1 and GIP receptors. The amylin system is a separate pathway — and this is exactly where Eloralintide acts.
AMYLIN RECEPTOR
Amylin receptors (AMY1–AMY3) are found in the area postrema and in the brainstem nuclei where AMY1–3 signalling is studied. Eloralintide activates them selectively.
SLOWER GASTRIC EMPTYING
In studies of amylin signalling, gastric emptying and post-meal glucagon are recorded as parameters.

Phase 1 — first data in humans
In the early study with repeated weekly administration, Eloralintide showed a dose-dependent reduction in body weight within 12 weeks, with a tolerability profile dominated by mild gastrointestinal discomfort.

Is Eloralintide a GLP-1 agonist?
