What is TB-500?

TB-500 (TB500, Thymosin Beta-4 fragment, Tβ4 fragment) is a synthetic peptide derived from the bioactive region of Thymosin Beta-4 — a natural protein of 43 amino acids, present in almost every cell and involved in tissue regeneration. TB-500 contains the key motif LKKTET, responsible for actin binding, thus concentrating the main cell-migration, angiogenesis and repair properties of the parent molecule.

Together with BPC-157, TB-500 is among the most studied peptides in the “regeneration and recovery” category. The two have complementary mechanisms — which is why they appear together in formulas such as BB20 or GLOW70.

It is studied in research for cell migration and differentiation, angiogenesis (formation of new blood vessels), repair of tissues and connective tissue, as well as anti-inflammatory actions. See the range in the all peptides category.

In the Peptology version, it is supplied as a lyophilised research-grade powder with purity > 99%, in two formats — 5 mg and 10 mg. It is intended exclusively for scientific, research and in vitro applications — it is not a medicine and is prohibited in sport (WADA).

Available formats: 5 mg and 10 mg

5 MG

Typical form

For validation experiments and protocols of usual size.

10 MG

Extended form

For longer courses and higher-consumption protocols.

Both forms have purity > 99% and a laboratory certificate for each batch. The choice depends on the scale of the protocol.

How TB-500 works — actin binding

ACTIN BINDING

Through the LKKTET motif, TB-500 binds monomeric G-actin (1:1) and regulates the G-actin ↔ F-actin balance — the basis of cytoskeleton dynamics and motility.

CELL MIGRATION

By regulating the actin cytoskeleton, it promotes the migration of cells (endothelial and progenitor) towards repair sites — a critical step of regeneration.

ANGIOGENESIS

In preclinical studies, TB-500 promotes the formation of new blood vessels (via VEGF) and has anti-inflammatory actions — supporting the oxygen supply to the tissue.

Why TB-500 and BPC-157 are “complementary”

TB-500 acts mainly on the cytoskeleton and cell migration (mobilising cells towards the target), while BPC-157 acts mainly on local angiogenesis and growth pathways. This very complementarity explains why they are often studied together.

Unlike the parent molecule (Thymosin Beta-4, 43 amino acids), TB-500 is a synthetic, smaller and more stable form, with good tissue distribution. The mechanisms come from preclinical studies and are not a guarantee of the result of a research product.

Properties and pharmacology

  • Origin: synthetic fragment of Thymosin Beta-4 (a natural protein of 43 amino acids)
  • Functional motif: the LKKTET sequence, the actin-binding region
  • Molecular target: monomeric G-actin (1:1 binding)
  • Studied actions: cell migration, angiogenesis (VEGF), repair, anti-inflammatory
  • Distribution: good tissue penetration thanks to its small size and stability
  • Status: research peptide, not approved; prohibited in sport (WADA)

Research

Tissue repair and cell migration

In vitro and animal studies have shown that Thymosin Beta-4 and its fragments promote the migration of endothelial cells and keratinocytes, accelerating wound healing in experimental models.

Angiogenesis and inflammation

In preclinical models, Thymosin Beta-4 promotes the formation of new vessels and regulates the inflammatory response, with investigation in cardiac, corneal and connective-tissue repair.

Data limitations

Most data come from preclinical studies; extensive clinical studies in humans confirming the efficacy and safety of TB-500 as it stands are lacking. It is also prohibited by WADA.

Note: Scientific reference based on preclinical data. It is not medical advice or a claim for the product.

Side effects and safety

The safety profile of TB-500 in humans is not fully characterised, as the data come mainly from preclinical studies. The reactions reported are generally mild.

Common

  • Injection-site reactions
  • Transient fatigue
  • Mild “brain fog” sensation / lethargy

Less common

  • Headache
  • Transient redness
  • Mild nausea

Precautions

  • Allergic reaction / hypersensitivity
  • Limited safety data in humans

Research warning: TB-500 is offered exclusively for scientific, research and in vitro purposes. It is not intended for human/veterinary use, or for the diagnosis, treatment or prevention of any condition, and does not constitute medical advice. TB-500 is prohibited in sport (WADA).

Difference from BPC-157?

They are complementary: TB-500 acts mainly on cell migration (the actin cytoskeleton), BPC-157 on local angiogenesis. That is why they appear together in BB20 .

Can it be combined with other peptides?

Yes. It combines excellently with BPC-157 , GH peptides (such as CJC-1295), as well as GHRP peptides for a stronger regenerative effect.

Research protocols

Important: Reference information for research purposes — not a recommendation for use or medical advice. TB-500 is not approved for human/veterinary use.

In the research literature, TB-500 protocols usually have a “loading” phase and a “maintenance” phase:

  • The loading phase generally lasts a few weeks, with more frequent administration
  • The maintenance phase uses a reduced frequency, because of the prolonged tissue distribution
  • Thanks to its stability and small size, TB-500 does not require daily administration in many protocols

TB-500 is often studied together with BPC-157 — a combination available ready-made in BB20. The schedules come from the research literature and do not constitute a recommendation for individual use.

Storage and reconstitution

It is supplied as a lyophilised powder in a vial (5 or 10 mg). Lyophilised, it is stable at room temperature, away from light/humidity. For reconstitution, the recommended solvent is Peptide Matrix Water™, which significantly extends the shelf life of the solution.

  • Before reconstitution: room temperature, away from light/moisture; up to 24 months.
  • With Peptide Matrix Water™ (recommended): refrigerator 2–8 °C up to 60 days — the best stability option.
  • With bacteriostatic water or sterile water: refrigerator 2–8 °C up to 28 days. No re-freezing.
  • Reconstitution: add the solvent slowly down the wall, without vigorous agitation.

Laboratory tip: Add Peptide Matrix Water™ to your basket — the recommended stabilising solvent for all reconstituted peptides.

Laboratory protocol — reconstitution, research protocols, storage

Standard protocol, applicable to all research peptides in the Peptology catalogue. For the concentration of the solution (mg/mL) per vial, use the peptide reconstitution calculator.

The 60-second protocol

2. RECONSTITUTION

The solvent is added to the wall of the vial, without agitation, then wait 10–15 minutes.

3. CALCULATION

Calculator → concentration in mg/mL

4. STORAGE

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water

Quick reference

Parameter Value
Form Lyophilised powder
Solvent Peptide Matrix Water
Volume for reconstitution 1–3 mL — 3 mL maximum (capacity of the standard vial)
Storage before reconstitution Room temperature, protected from direct light (refrigeration is not mandatory)
After reconstitution Up to 60 days at +2 °C to +8 °C with Peptide Matrix Water

What you will need

Precision pipette

Calibrated, 100–1000 µL — for accurate measurement of the volume of solvent added to the vial.

70% alcohol pads

Two pads — to disinfect the peptide vial and the solvent vial.

Peptide Matrix Water

Stabilised solvent for lyophilised peptides. Why this one.

Five rules for reconstitution

✅ Correct ❌ Wrong
Disinfect the stoppers and the work surface with 70% alcohol, work with gloves Working without disinfection
The solvent is added slowly down the wall of the vial Direct jet onto the powder — foaming and denaturation
Light swirling between the palms Shaking — foam, on which the peptides denature
Wait 10–15 minutes at room temperature Mechanical mixing or heating
Visual inspection: clear, without particles Using a cloudy or coloured solution

Calculating the concentration

⚠️ Maximum 3 mL of solvent. The standard vial does not allow a larger volume — do not exceed it during reconstitution.

  • Concentration (mg/mL) = mg in the vial ÷ mL of solvent
  • Volume per sample (mL) = amount per sample (mg) ÷ concentration (mg/mL)
  • 1 mg/mL = 1 µg/µL · 1 mL = 1000 µL

Reconstitution per vial — solvent: Peptide Matrix Water. Concentration after addition (1 mg/mL = 1 µg/µL):

Vial+ 1 mL+ 2 mL+ 3 mL
5 mg5 mg/mL2.5 mg/mL1.67 mg/mL
10 mg10 mg/mL5 mg/mL3.33 mg/mL

👉 For any vial and solvent volume — use the calculator. It calculates the concentration (mg/mL, µg/µL) and the volume per sample (mL, µL).

Storage

BEFORE RECONSTITUTION

Room temperature, protected from direct light, in the original packaging. Refrigeration is not mandatory.

AFTER RECONSTITUTION

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water. Discard the remainder after this period.

FREEZING

Not recommended. Freeze/thaw cycles degrade the structure of the peptide.

Four mistakes to avoid

1. Shaking for “faster dissolution”

Foam = denaturation. Only gentle swirling between the palms.

2. Unsuitable solvent

For the stability of the peptide structure, use only a sterile, purpose-designed solvent — Peptide Matrix Water is recommended.

3. Ignoring slight cloudiness

If it is not completely clear = the sample does not meet the criteria for work.

4. Removing it from the refrigerator “for an hour or two”

Short exposures add up and jeopardise stability.
