What is PT-141?

PT-141 (Bremelanotide) is a peptide agonist of the melanocortin receptors, a derivative of Melanotan II. Unlike Melanotan II (which is mainly used for pigmentation), PT-141 acts selectively on the pathways of libido and sexual arousal, through the central nervous system — not through peripheral vascular mechanisms.

PT-141 is unique in that it acts on the brain (hypothalamus and limbic regions), stimulating sexual desire and arousal in a neurogenic way — a mechanism different from that of PDE5 inhibitors (which act on blood flow).

It has been studied clinically (Phase III) in hypoactive sexual desire disorder (HSDD) in premenopausal women. See the range in the all peptides category.

In the Peptology version, it is supplied as a lyophilised powder for research use with purity > 99%, in a 10 mg format. It is NOT an approved medicine — it is intended exclusively for scientific, research and in vitro applications.

How PT-141 acts — melanocortin receptors

MC4R / MC3R RECEPTORS

It selectively activates the melanocortin receptors MC4R and MC3R in the hypothalamus and limbic regions — the centres of motivation and reward.

DOPAMINE RELEASE

It promotes the release of neurotransmitters such as dopamine, which play a central role in desire and pleasure — a neurogenic, not a vascular, pathway.

CENTRAL ACTION

The action takes place in the central nervous system, not in the vessels — a fact that differentiates it from PDE5 inhibitors and makes it complementary.

Why “short half-life, prolonged action”

PT-141 has a half-life of approximately 2.7 hours, but the subjective effect lasts much longer. In clinical studies, the onset of arousal was measured at 45–90 minutes after administration, lasting 6–8 hours (occasionally up to 8–12). This suggests that the activation of melanocortin receptors in the brain produces effects that exceed the presence of the peptide in the blood.

The mechanisms come from clinical studies with the approved medicine and are not a guarantee of the result of a research product.

Properties and pharmacokinetics

  • Molecule type: melanocortin agonist peptide (metabolite of Melanotan II)
  • Targets: MC4R and MC3R (hypothalamus, limbic regions)
  • Route of action: central (CNS), neurogenic — not vascular
  • Half-life: approximately 2.7 hours (range 1.9–4.0)
  • Onset / duration: onset 45–90 minutes; duration 6–8 hours (up to 8–12)
  • Regulatory history: Phase III clinical studies in HSDD

Clinical studies

Hypoactive sexual desire disorder (HSDD)

In Phase III clinical studies, bremelanotide improved sexual desire and reduced the associated distress in premenopausal women with HSDD.

A central, not vascular, mechanism

Unlike PDE5 inhibitors, PT-141 acts through MC4Rs in the brain — a fact that explains the effect on desire (not only on the physical response) and the research interest in both sexes.

Research directions

The role of MC4R in sexual desire continues to be studied, in contexts that go beyond the original approved indication.

Research protocols

Important: Reference information — not a recommendation for use or medical advice. Research-grade PT-141 is not an approved medicine.

In a clinical context, bremelanotide was administered subcutaneously, on demand:

  • Subcutaneous administration, approximately 45 minutes before the targeted moment
  • Onset appears in 45–90 minutes; the duration extends to 6–8 hours
  • In the approved form, a maximum of one dose per 24 hours and a limited number per month is recommended

The schedules come from clinical use and are not a recommendation for individual use.

Storage and reconstitution

Supplied as a lyophilised powder in a 10 mg vial. Lyophilised, it is stable at room temperature, away from light/moisture. For reconstitution, the recommended solvent is Peptide Matrix Water™, which significantly extends the life of the solution.

  • Before reconstitution: room temperature, away from light/moisture; up to 24 months.
  • With Peptide Matrix Water™ (recommended): refrigerator 2–8 °C up to 60 days — the best stability option.
  • With bacteriostatic water: refrigerator 2–8 °C for up to 28 days. Do not re-freeze.
  • Reconstitution: add the solvent slowly down the wall, without vigorous agitation.

Laboratory tip: Add Peptide Matrix Water™ to your basket — the recommended stabilising solvent for all reconstituted peptides.

Laboratory protocol — reconstitution, research protocols, storage

Standard protocol, applicable to all research peptides in the Peptology catalogue. For the concentration of the solution (mg/mL) per vial, use the peptide reconstitution calculator.

The 60-second protocol

2. RECONSTITUTION

The solvent is added to the wall of the vial, without agitation, then wait 10–15 minutes.

3. CALCULATION

Calculator → concentration in mg/mL

4. STORAGE

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water

Quick reference

Parameter Value
Form Lyophilised powder
Solvent Peptide Matrix Water
Volume for reconstitution 1–3 mL — 3 mL maximum (capacity of the standard vial)
Storage before reconstitution Room temperature, protected from direct light (refrigeration is not mandatory)
After reconstitution Up to 60 days at +2 °C to +8 °C with Peptide Matrix Water

What you will need

Precision pipette

Calibrated, 100–1000 µL — for accurate measurement of the volume of solvent added to the vial.

70% alcohol pads

Two pads — to disinfect the peptide vial and the solvent vial.

Peptide Matrix Water

Stabilised solvent for lyophilised peptides. Why this one.

Five rules for reconstitution

✅ Correct ❌ Wrong
Disinfect the stoppers and the work surface with 70% alcohol, work with gloves Working without disinfection
The solvent is added slowly down the wall of the vial Direct jet onto the powder — foaming and denaturation
Light swirling between the palms Shaking — foam, on which the peptides denature
Wait 10–15 minutes at room temperature Mechanical mixing or heating
Visual inspection: clear, without particles Using a cloudy or coloured solution

Calculating the concentration

⚠️ Maximum 3 mL of solvent. The standard vial does not allow a larger volume — do not exceed it during reconstitution.

  • Concentration (mg/mL) = mg in the vial ÷ mL of solvent
  • Volume per sample (mL) = amount per sample (mg) ÷ concentration (mg/mL)
  • 1 mg/mL = 1 µg/µL · 1 mL = 1000 µL

Reconstitution per vial — solvent: Peptide Matrix Water. Concentration after addition (1 mg/mL = 1 µg/µL):

Vial+ 1 mL+ 2 mL+ 3 mL
10 mg10 mg/mL5 mg/mL3.33 mg/mL

👉 For any vial and solvent volume — use the calculator. It calculates the concentration (mg/mL, µg/µL) and the volume per sample (mL, µL).

Storage

BEFORE RECONSTITUTION

Room temperature, protected from direct light, in the original packaging. Refrigeration is not mandatory.

AFTER RECONSTITUTION

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water. Discard the remainder after this period.

FREEZING

Not recommended. Freeze/thaw cycles degrade the structure of the peptide.

Four mistakes to avoid

1. Shaking for “faster dissolution”

Foam = denaturation. Only gentle swirling between the palms.

2. Unsuitable solvent

For the stability of the peptide structure, use only a sterile, purpose-designed solvent — Peptide Matrix Water is recommended.

3. Ignoring slight cloudiness

If it is not completely clear = the sample does not meet the criteria for work.

4. Removing it from the refrigerator “for an hour or two”

Short exposures add up and jeopardise stability.
