What is Vilon?

Vilon is a synthetic Lys-Glu dipeptide (lysine-glutamic acid, also known as KE), one of the short peptide bioregulators of the Khavinson school (Institute of Bioregulation and Gerontology, St Petersburg). It is cited as one of the shortest peptides with documented biological activity — only two amino acids.

Bioregulators are a class of short peptides studied for tissue-specific regulation of gene expression. Vilon is the thymic/immune representative of this family and is studied mainly for regulation of immune function and in the context of longevity. The same group includes Epithalon (a telomeric/pineal bioregulator) and thymic peptides such as Thymulin.

At Peptology it is supplied as a lyophilised research peptide with purity above 99% (see certificates), with a price in Cyprus and pay on delivery. Exclusively for research purposes; it is not a medicine. See also the related bioregulators Epithalon and Thymulin.

How Vilon is studied — mechanism

In the Khavinson school literature, Vilon is described as a bioregulator acting at the level of gene expression in immune cells. The three axes below summarise the main directions of study of the dipeptide.

IMMUNE REGULATION

Studied as a thymic/immune bioregulator, linked to the function and maturation of T lymphocytes.

GENE EXPRESSION

Regulation of gene expression through “chromatin decondensation” is described — activation of “silent” genes in aged cells.

GERONTOLOGY

Examined in the context of bioregulation and restoration of functions during ageing.

The concept of peptide bioregulation

The hypothesis of the Khavinson school is that very short peptides can enter the cell and the nucleus and bind to specific regions of DNA, influencing which genes are active. During ageing some genes become “silent” through chromatin condensation; according to this concept, bioregulators such as Vilon can “open” such regions and restore their expression — a mechanism studied mainly in immune and thymic cells.

Why the thymus

The thymus is the organ where T lymphocytes mature, and it shrinks progressively with age (atrophy), which is linked to a weakening of the immune response. Vilon is studied precisely as a thymic bioregulator — in the context of maintaining immune function during ageing.

Khavinson's bioregulators

The family of short peptide bioregulators was developed in St Petersburg and includes tissue-specific peptides, each directed at a specific organ or system. Below is Vilon's place among them.

VILON (KE)

Thymic/immune bioregulator — Lys-Glu dipeptide.

EPITHALON

Pineal/telomeric bioregulator — AEDG tetrapeptide.

THYMIC PEPTIDES

Thymulin, Thymosin — immune regulation and neuroimmune regulation.

Scientific basis and availability of data

Research by the Khavinson school

Most of the research on Vilon comes from the group of Prof. Khavinson and collaborators. It describes effects on the maturation and function of T lymphocytes, on immune regulation and on ageing markers in animal and cell models, as well as interactions with components of the NF-κB pathway.

Limitations of the evidence

Since much of the data comes from a single research group, independent replication is limited. This does not diminish the research value of Vilon, but it requires that efficacy data be interpreted with this reservation — and it is the reason it is offered only for research purposes.

The information is a scientific reference and concerns mainly research by the Khavinson group. It is not a claim of results or a recommendation for use.

Vilon is available in: 20 mg.

Research protocols

Important: The information below summarises ranges observed among researchers and is not a recommendation for use, medical advice or an application instruction.

  • The 20 mg vial is usually reconstituted with ~3 mL of liquid (~6.67 mg/mL).
  • In protocols, bioregulators are applied in short cycles (e.g. ~5–10 days) within a longer period.
  • The concentration is determined by the reconstitution volume and the aims of the research.

Storage and reconstitution

  • Before reconstitution: lyophilised at −20 °C (−80 °C for long-term storage), in the dark and dry.
  • After reconstitution: in the refrigerator at 2–8 °C, use within ~a week, or divide into portions and freeze.
  • Reconstitution: add the liquid slowly down the wall of the vial; swirl gently, do not shake.

For reconstitution we recommend Peptide Matrix Water™ — a stabilising solution for reconstituted research peptides; alternatively, bacteriostatic water.

Laboratory protocol — reconstitution, research protocols, storage

Standard protocol, applicable to all research peptides in the Peptology catalogue. For the concentration of the solution (mg/mL) per vial, use the peptide reconstitution calculator.

The 60-second protocol

2. RECONSTITUTION

The solvent is added to the wall of the vial, without agitation, then wait 10–15 minutes.

3. CALCULATION

Calculator → concentration in mg/mL

4. STORAGE

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water

Quick reference

Parameter Value
Form Lyophilised powder
Solvent Peptide Matrix Water
Volume for reconstitution 1–3 mL — 3 mL maximum (capacity of the standard vial)
Storage before reconstitution Room temperature, protected from direct light (refrigeration is not mandatory)
After reconstitution Up to 60 days at +2 °C to +8 °C with Peptide Matrix Water

What you will need

Precision pipette

Calibrated, 100–1000 µL — for accurate measurement of the volume of solvent added to the vial.

70% alcohol pads

Two pads — to disinfect the peptide vial and the solvent vial.

Peptide Matrix Water

Stabilised solvent for lyophilised peptides. Why this one.

Five rules for reconstitution

✅ Correct ❌ Wrong
Disinfect the stoppers and the work surface with 70% alcohol, work with gloves Working without disinfection
The solvent is added slowly down the wall of the vial Direct jet onto the powder — foaming and denaturation
Light swirling between the palms Shaking — foam, on which the peptides denature
Wait 10–15 minutes at room temperature Mechanical mixing or heating
Visual inspection: clear, without particles Using a cloudy or coloured solution

Calculating the concentration

⚠️ Maximum 3 mL of solvent. The standard vial does not allow a larger volume — do not exceed it during reconstitution.

  • Concentration (mg/mL) = mg in the vial ÷ mL of solvent
  • Volume per sample (mL) = amount per sample (mg) ÷ concentration (mg/mL)
  • 1 mg/mL = 1 µg/µL · 1 mL = 1000 µL

Reconstitution per vial — solvent: Peptide Matrix Water. Concentration after addition (1 mg/mL = 1 µg/µL):

Vial+ 1 mL+ 2 mL+ 3 mL
20 mg20 mg/mL10 mg/mL6.67 mg/mL

👉 For any vial and solvent volume — use the calculator. It calculates the concentration (mg/mL, µg/µL) and the volume per sample (mL, µL).

Storage

BEFORE RECONSTITUTION

Room temperature, protected from direct light, in the original packaging. Refrigeration is not mandatory.

AFTER RECONSTITUTION

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water. Discard the remainder after this period.

FREEZING

Not recommended. Freeze/thaw cycles degrade the structure of the peptide.

Four mistakes to avoid

1. Shaking for “faster dissolution”

Foam = denaturation. Only gentle swirling between the palms.

2. Unsuitable solvent

For the stability of the peptide structure, use only a sterile, purpose-designed solvent — Peptide Matrix Water is recommended.

3. Ignoring slight cloudiness

If it is not completely clear = the sample does not meet the criteria for work.

4. Removing it from the refrigerator “for an hour or two”

Short exposures add up and jeopardise stability.
