What is Tesamorelin?

Tesamorelin (TH9507) is a stabilised synthetic analogue of GHRH (growth hormone-releasing hormone) — a peptide of 44 amino acids with a modification at the N-terminus that protects it from rapid degradation. It stimulates the pituitary to secrete growth hormone (GH) in a natural, pulsatile manner. It is the most studied GHRH analogue for reduction of visceral (abdominal) fat.

Unlike shorter-lived GHRH analogues such as CJC-1295 without DAC, Tesamorelin has a stronger and more stable action on the GH/IGF-1 axis.

It is studied in research for reduction of visceral adipose tissue (VAT), raising IGF-1 levels, improving body composition and, in some works, for cognitive endpoints. See the whole range in the all peptides category.

In the Peptology version, it is supplied as a lyophilised research-grade powder with purity > 99%, in two formats — 10 mg and 30 mg. It is intended exclusively for scientific, research and in vitro applications — it is not a medicine or a supplement and is prohibited in sport (WADA).

Available formats: 10 mg and 30 mg

10 MG

Typical form

For validation experiments and protocols of usual size.

30 MG

Extended form

For longer courses and higher-consumption protocols.

⚠ Important for Tesamorelin 30 mg: Because of its high concentration, it is a special, highly concentrated peptide. It is perfectly normal for the solution to look slightly milky/off-white after reconstitution. It is NOT normal for the solution to be yellow or brown, or to contain visible particles — in that case do not use it.

Both forms have purity > 99% and a laboratory certificate for each batch. The choice depends on the scale of the protocol.

How Tesamorelin works — the GHRH → GH → IGF-1 axis

PITUITARY STIMULATION

As a GHRH analogue, it binds to the pituitary GHRH receptors and stimulates growth hormone secretion — preserving natural pulsatility.

IGF-1 INCREASE

The increased GH stimulates the liver to produce IGF-1, the main mediator of the anabolic and metabolic actions of the axis.

VAT LIPOLYSIS

Growth hormone promotes lipolysis, with a particular effect on visceral fat — the main studied endpoint of Tesamorelin.

Why a “stabilised” analogue

Natural GHRH is degraded rapidly in plasma. Tesamorelin carries a modification (trans-3-hexenoyl) at the N-terminus that increases stability and duration of action, while retaining the physiological, pulsatile character of GH secretion — unlike the continuous stimulation caused by some other molecules.

Because it acts upstream on the axis (stimulating the pituitary itself), the natural negative-feedback mechanism is preserved — a theoretical safety advantage over exogenous GH administration. The mechanisms come from clinical and preclinical studies.

Research

Reduction of visceral fat

In randomised controlled clinical studies, Tesamorelin significantly reduced visceral adipose tissue in people with accumulation of abdominal fat, with a corresponding improvement in body-composition markers.

IGF-1 and metabolic markers

Administration raises IGF-1 levels, with studied effects on triglycerides and other metabolic markers. Monitoring of IGF-1 is a central element in research protocols.

Data limitations

Although there are strong clinical data in the approved population, use outside it is not fully characterised. Tesamorelin is also prohibited by WADA.

Side effects and safety

From the clinical data, the reactions reported are mainly related to the increased GH/IGF-1 action and to injection-site reactions.

Common

  • Injection-site reactions
  • Arthralgia / myalgia
  • Oedema / fluid retention

Less common

  • Paraesthesia / numbness
  • Carpal tunnel syndrome
  • Changes in glucose

Precautions

  • Allergic reaction / hypersensitivity
  • Contraindicated in active malignancy (IGF-1 axis)
  • Caution regarding glucose intolerance

Because the GH/IGF-1 axis is related to cell proliferation, there is a precaution in states of active malignancy. Scientific reference, not instructions for use.

How does it differ from CJC-1295?

Both are GHRH analogues. Tesamorelin has strong clinical data on visceral fat; CJC-1295 with DAC has a much longer duration of action, while CJC without DAC gives a short, pulsatile stimulus.

Can it be combined with other peptides?

Yes. It is suitable for combination with CJC-1295, Ipamorelin or BPC-157.

Research protocols

Important: Reference information for research purposes — not a recommendation for use or medical advice.

In the clinical literature, Tesamorelin is generally administered on a daily schedule, because of the nature of the GHRH axis:

  • It is usually administered once a day, to maintain steady stimulation of the axis
  • Evening administration is studied for alignment with natural nocturnal GH secretion
  • IGF-1 is often monitored as a marker of response and safety

GHRH analogues are sometimes studied in combination with GH-releasing peptides (GHRPs). The schedules come from the literature and do not constitute a recommendation for individual use.

Storage and reconstitution

It is supplied as a lyophilised powder in a vial (10 or 30 mg). Lyophilised, it is stable when refrigerated, away from light/humidity. For reconstitution, the recommended solvent is Peptide Matrix Water™, which significantly extends the shelf life of the solution.

  • Before reconstitution: preferably refrigerated at 2–8 °C, away from light; stable for an extended period.
  • With Peptide Matrix Water™ (recommended): refrigerator 2–8 °C up to 60 days — the best stability option.
  • With bacteriostatic water or sterile water: refrigerator 2–8 °C up to 28 days. No re-freezing.
  • Reconstitution: add the solvent slowly down the wall, without vigorous agitation.

Laboratory tip: Add Peptide Matrix Water™ to your basket — the recommended stabilising solvent for all reconstituted peptides.

Laboratory protocol — reconstitution, research protocols, storage

Standard protocol, applicable to all research peptides in the Peptology catalogue. For the concentration of the solution (mg/mL) per vial, use the peptide reconstitution calculator.

The 60-second protocol

2. RECONSTITUTION

The solvent is added to the wall of the vial, without agitation, then wait 10–15 minutes.

3. CALCULATION

Calculator → concentration in mg/mL

4. STORAGE

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water

Quick reference

Parameter Value
Form Lyophilised powder
Solvent Peptide Matrix Water
Volume for reconstitution 1–3 mL — 3 mL maximum (capacity of the standard vial)
Storage before reconstitution Room temperature, protected from direct light (refrigeration is not mandatory)
After reconstitution Up to 60 days at +2 °C to +8 °C with Peptide Matrix Water

What you will need

Precision pipette

Calibrated, 100–1000 µL — for accurate measurement of the volume of solvent added to the vial.

70% alcohol pads

Two pads — to disinfect the peptide vial and the solvent vial.

Peptide Matrix Water

Stabilised solvent for lyophilised peptides. Why this one.

Five rules for reconstitution

✅ Correct ❌ Wrong
Disinfect the stoppers and the work surface with 70% alcohol, work with gloves Working without disinfection
The solvent is added slowly down the wall of the vial Direct jet onto the powder — foaming and denaturation
Light swirling between the palms Shaking — foam, on which the peptides denature
Wait 10–15 minutes at room temperature Mechanical mixing or heating
Visual inspection: clear, without particles Using a cloudy or coloured solution

Calculating the concentration

⚠️ Maximum 3 mL of solvent. The standard vial does not allow a larger volume — do not exceed it during reconstitution.

  • Concentration (mg/mL) = mg in the vial ÷ mL of solvent
  • Volume per sample (mL) = amount per sample (mg) ÷ concentration (mg/mL)
  • 1 mg/mL = 1 µg/µL · 1 mL = 1000 µL

Reconstitution per vial — solvent: Peptide Matrix Water. Concentration after addition (1 mg/mL = 1 µg/µL):

Vial+ 1 mL+ 2 mL+ 3 mL
10 mg10 mg/mL5 mg/mL3.33 mg/mL
30 mg30 mg/mL15 mg/mL10 mg/mL

👉 For any vial and solvent volume — use the calculator. It calculates the concentration (mg/mL, µg/µL) and the volume per sample (mL, µL).

Storage

BEFORE RECONSTITUTION

Room temperature, protected from direct light, in the original packaging. Refrigeration is not mandatory.

AFTER RECONSTITUTION

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water. Discard the remainder after this period.

FREEZING

Not recommended. Freeze/thaw cycles degrade the structure of the peptide.

Four mistakes to avoid

1. Shaking for “faster dissolution”

Foam = denaturation. Only gentle swirling between the palms.

2. Unsuitable solvent

For the stability of the peptide structure, use only a sterile, purpose-designed solvent — Peptide Matrix Water is recommended.

3. Ignoring slight cloudiness

If it is not completely clear = the sample does not meet the criteria for work.

4. Removing it from the refrigerator “for an hour or two”

Short exposures add up and jeopardise stability.
