What is CJC-1295?

CJC-1295 without DAC (also known as Modified GRF 1-29, Mod GRF 1-29) is a synthetic, short-acting GHRH analogue — of the hormone that releases growth hormone. It is a modified fragment of GRF (1-29) with four amino acid substitutions, which give it resistance to degradation by the peptidase DPP-4 and other plasma proteases, while retaining the short, physiological action profile.

The designation “without DAC” is critical. DAC (Drug Affinity Complex) is an albumin-binding group that, in the “with DAC” version, extends the half-life to days and maintains a steady, raised GH baseline. The version without DAC does not have this group — its half-life is ~30 minutes, producing short, pulsatile GH peaks that mimic more faithfully the natural secretion rhythm from the pituitary.

It acts on the GHRH receptor in the pituitary. This distinguishes it from the GHRP peptides / ghrelin mimetics (such as GHRP-2 or Hexarelin), which act on the separate GHS-R1a receptor. As these are two different pathways, in studies they are combined for a synergistic effect — hence the popularity of the combination with a GHRP / Ipamorelin. It is a research peptide with no approval as a medicine; it is studied in the GH/IGF-1 axis, regeneration, body composition and lipolysis.

At Peptology it is supplied as a lyophilised peptide with purity above 99% (HPLC/MS, CoA per batch), in a 5 mg pack. Exclusively for research and in vitro applications. See also the peptides for regeneration and anabolism.

Available pack: 5 mg

CJC-1295 without DAC is available in a lyophilised 5 mg form, covering the typical scales of research work — from individual in vitro runs to larger GH-axis protocols, including schedules in combination with GHRP peptides.

5 MG

Standard pack

A lyophilised GHRH analogue with purity > 99%, for studies of the GH/IGF-1 axis, regeneration and schedules in combination with GHRP.

COMBINATION

CJC-1295 + Ipamorelin

A ready-made blend of GHRH + GH secretagogue in one vial — for protocols that study the synergy of the two receptors.

The 5 mg pack has purity > 99% and a laboratory certificate for the specific batch. The choice of quantity depends on the scale of the protocol, not on the concentration of the active substance.

How it acts — stimulation of the GHRH axis

CJC-1295 without DAC stimulates the GHRH receptor in the pituitary and causes a short, pulsatile GH peak. The three axes below determine its profile.

GHRH RECEPTOR

It binds the GHRH receptor, activating GH synthesis and release via cAMP — GH peak usually within 15–30 minutes.

SHORT PROFILE (WITHOUT DAC)

The absence of an albumin-binding group (DAC) means a half-life of ~30 minutes — short, pulsatile GH peaks instead of a steady baseline.

SYNERGY WITH GHRP

The GHRH receptor acts in parallel with GHS-R1a (GHRP/ghrelin). The combination is associated with a 3–5-fold increase in GH release.

Without DAC vs with DAC — the difference

The version with DAC has an albumin-binding group that extends the half-life to days and maintains a steady, raised GH/IGF-1 baseline. The version without DAC (this product) acts briefly (~30 minutes) and causes isolated, physiological GH pulses. Because the GHRH and GHS-R1a (GHRP/ghrelin) pathways are independent, in protocols the GHRH analogue is combined with a secretagogue — e.g. GHRP-2, Hexarelin or Ipamorelin. Data from pharmacological studies, not a claim about the research-grade product.

Pharmacokinetics

  • Half-life: ~30 minutes (absence of a DAC group)
  • GH peak: usually 15–30 minutes after subcutaneous administration
  • Action profile: short, pulsatile — mimics the natural GH rhythm
  • Site of action: GHRH receptor in the pituitary (cAMP pathway)
  • Enzymatic stability: 4 amino acid substitutions → resistance to DPP-4
  • Schedule: subcutaneous, often in combination with a GHRP

CJC-1295 without DAC vs Sermorelin vs GHRP-2

Sermorelin is another GHRH analogue (same receptor), while GHRP-2 is a ghrelin mimetic at GHS-R1a. Scientific comparison data.

Clinical and preclinical data

CJC-1295 is a research molecule — it has not gone through a phase III approval programme. Most controlled clinical data relate to the form with DAC: in early studies in healthy adults, a dose-dependent increase in GH and IGF-1 was documented after subcutaneous administration, with good tolerability. The form without DAC (Mod GRF 1-29) causes short GH peaks (15–30 minutes) that mimic the endogenous rhythm — a tool for studies of the pulsatility of the GH axis.

The combination with a GHRP secretagogue is associated with 3–5-fold higher GH release (separate receptors). The information is scientific and informative; it is not medical advice or a claim about the research-grade product.

All information comes from the pharmacological and research literature and is provided for scientific and informative purposes; it is not a claim about the action of the research-grade product offered by Peptology.

Is it combined with a GHRP?

Yes — a classic research schedule. CJC-1295 acts on GHRH-R and the GHRPs ( GHRP-2 , Hexarelin , Ipamorelin) on GHS-R1a, with 3–5× higher GH release. Ready-made: CJC-1295 + Ipamorelin .

How does it differ from Sermorelin?

Both are GHRH analogues at the same receptor, but CJC-1295 (Mod GRF 1-29) has four amino acid substitutions that increase resistance to DPP-4 — a slightly more stable profile than Sermorelin.

Laboratory protocol — reconstitution, research protocols, storage

Standard protocol, applicable to all research peptides in the Peptology catalogue. For the concentration of the solution (mg/mL) per vial, use the peptide reconstitution calculator.

The 60-second protocol

2. RECONSTITUTION

The solvent is added to the wall of the vial, without agitation, then wait 10–15 minutes.

3. CALCULATION

Calculator → concentration in mg/mL

4. STORAGE

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water

Quick reference

Parameter Value
Form Lyophilised powder
Solvent Peptide Matrix Water
Volume for reconstitution 1–3 mL — 3 mL maximum (capacity of the standard vial)
Storage before reconstitution Room temperature, protected from direct light (refrigeration is not mandatory)
After reconstitution Up to 60 days at +2 °C to +8 °C with Peptide Matrix Water

What you will need

Precision pipette

Calibrated, 100–1000 µL — for accurate measurement of the volume of solvent added to the vial.

70% alcohol pads

Two pads — to disinfect the peptide vial and the solvent vial.

Peptide Matrix Water

Stabilised solvent for lyophilised peptides. Why this one.

Five rules for reconstitution

✅ Correct ❌ Wrong
Disinfect the stoppers and the work surface with 70% alcohol, work with gloves Working without disinfection
The solvent is added slowly down the wall of the vial Direct jet onto the powder — foaming and denaturation
Light swirling between the palms Shaking — foam, on which the peptides denature
Wait 10–15 minutes at room temperature Mechanical mixing or heating
Visual inspection: clear, without particles Using a cloudy or coloured solution

Calculating the concentration

⚠️ Maximum 3 mL of solvent. The standard vial does not allow a larger volume — do not exceed it during reconstitution.

  • Concentration (mg/mL) = mg in the vial ÷ mL of solvent
  • Volume per sample (mL) = amount per sample (mg) ÷ concentration (mg/mL)
  • 1 mg/mL = 1 µg/µL · 1 mL = 1000 µL

Reconstitution per vial — solvent: Peptide Matrix Water. Concentration after addition (1 mg/mL = 1 µg/µL):

Vial+ 1 mL+ 2 mL+ 3 mL
5 mg5 mg/mL2.5 mg/mL1.67 mg/mL

👉 For any vial and solvent volume — use the calculator. It calculates the concentration (mg/mL, µg/µL) and the volume per sample (mL, µL).

Storage

BEFORE RECONSTITUTION

Room temperature, protected from direct light, in the original packaging. Refrigeration is not mandatory.

AFTER RECONSTITUTION

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water. Discard the remainder after this period.

FREEZING

Not recommended. Freeze/thaw cycles degrade the structure of the peptide.

Four mistakes to avoid

1. Shaking for “faster dissolution”

Foam = denaturation. Only gentle swirling between the palms.

2. Unsuitable solvent

For the stability of the peptide structure, use only a sterile, purpose-designed solvent — Peptide Matrix Water is recommended.

3. Ignoring slight cloudiness

If it is not completely clear = the sample does not meet the criteria for work.

4. Removing it from the refrigerator “for an hour or two”

Short exposures add up and jeopardise stability.
