What is HCG?

HCG (human chorionic gonadotropin) is a glycoprotein hormone that acts as an analogue of luteinising hormone (LH). It binds with high affinity to the LH/HCG receptor (LHCGR) and triggers the same pathways as natural LH — which is why it is used in research on the hypothalamic-pituitary-gonadal (HPG) axis, fertility and testosterone production.

Unlike molecules that act upstream in the HPG axis — such as Kisspeptin-10 (in the hypothalamus) or Gonadorelin (in the pituitary) — HCG acts directly on the gonads, stimulating the Leydig cells of the testis to produce testosterone.

It is studied in research for steroidogenesis (testosterone production), the maintenance of testicular function and spermatogenesis and the HPG axis. See the range in the all peptides category.

In the Peptology version, HCG is supplied as a lyophilised powder for research use in a 5000 IU format, with a batch certificate. It is intended exclusively for scientific, research and in vitro applications — it is not a medicine and is not approved for human/veterinary use.

How HCG acts — an LH analogue

LH/HCG RECEPTOR

HCG binds with high affinity to the LHCGR receptor (G-protein-coupled) on Leydig cells, mimicking the action of luteinising hormone.

cAMP / PKA

Activation increases intracellular cAMP and PKA, triggering steroidogenesis through the StAR protein and the enzymes CYP11A1 and CYP17A1.

INTRATESTICULAR TESTOSTERONE

The result is testosterone production in the testis, where the concentration is 50–100× higher than in the blood — critical for spermatogenesis.

Why HCG maintains testicular function

The testosterone concentration inside the testis is 50–100 times higher than in the blood, and this gradient is essential for the development of sperm. Clinical studies (Fertility and Sterility, The Journal of Urology) show that HCG, as an adjunctive therapy, can maintain sperm count in men receiving exogenous testosterone — which otherwise suppresses LH production.

Recent research notes that, unlike LH (which also activates survival/proliferation signals), HCG mainly triggers steroidogenic signals. The mechanisms come from studies and are not a guarantee of the result of a research product.

Properties and pharmacology

  • Molecule type: glycoprotein hormone (α and β subunits)
  • Target: the LH/HCG receptor (LHCGR), G-protein-coupled
  • Signalling pathway: Gs → cAMP → PKA → StAR, CYP11A1, CYP17A1
  • Main action: steroidogenesis — testosterone production in Leydig cells
  • Particularity: a longer half-life than LH (due to glycosylation)
  • Status: a research hormone in this context; prohibited in sport for men (WADA)

Clinical studies

Maintenance of spermatogenesis

Studies show that HCG, as an adjunctive therapy, can maintain sperm production in men receiving exogenous testosterone — by maintaining intratesticular testosterone, which exogenous testosterone suppresses.

Hypogonadism and fertility

HCG is studied in hypogonadotropic hypogonadism and in various approaches to optimising male fertility, including non-obstructive azoospermia (andrology literature).

Dose and safety of Leydig cells

Recent research notes that exposure to high doses of HCG can cause endoplasmic reticulum stress and apoptosis in Leydig cells — a finding that draws attention to appropriate dose and duration ranges.

Side effects and safety

The safety profile of HCG is well characterised from clinical use. Many reactions are local or related to the rise in testosterone and oestrogen.

Common

  • Injection-site reactions
  • Headache
  • Mood changes

Less common

  • Gynaecomastia (raised oestrogen)
  • Fluid retention
  • Acne

Precautions

  • Allergic reaction / hypersensitivity
  • Leydig apoptosis at high, prolonged doses
  • Aromatisation → raised oestrogen

Part of the testosterone produced under HCG is converted (aromatised) into oestrogens, which explains some of the effects. Scientific reference, not instructions for use.

Research warning: HCG is offered exclusively for scientific, research and in vitro purposes. It is not intended for human/veterinary use, or for the diagnosis, treatment or prevention of any condition, and is not medical advice. HCG is prohibited in sporting competitions (for men) by WADA.

How does it differ from Gonadorelin and Kisspeptin-10?

They act at different levels of the HPG axis: Kisspeptin-10 in the hypothalamus, Gonadorelin in the pituitary, and HCG directly on the gonads. HCG is the only one that directly stimulates testosterone production.

HCG is available in: 5000 IU.

Research protocols

Important: Reference information from the clinical literature — not a recommendation for use or medical advice. HCG is not approved for human use in this context.

In the clinical literature, HCG was administered subcutaneously, at relatively small doses and moderate frequency:

  • The usual protocols describe 500–1000 IU subcutaneously, 2–3 times a week
  • This dose maintains testicular function without an excessive rise in serum testosterone
  • High, prolonged doses are avoided because of the stress on Leydig cells

A reconstituted 5000 IU vial covers many administrations at these doses. The schedules come from the research literature and are not a recommendation for individual use.

Storage and reconstitution

HCG is supplied as a lyophilised powder in a 5000 IU vial. In lyophilised form it is stable, away from light/moisture. After reconstitution, HCG must be stored in the refrigerator; for reconstitution, the recommended solvent is Peptide Matrix Water™ (alternatively, bacteriostatic water), which extends the shelf life of the solution.

  • Before reconstitution: preferably in the refrigerator; away from light/moisture.
  • With Peptide Matrix Water™ (recommended): refrigerator 2–8 °C up to 60 days — the best stability option.
  • With bacteriostatic water: refrigerator 2–8 °C, usually for up to ~28–30 days. Do not re-freeze.
  • Reconstitution: add the solvent slowly down the wall, without vigorous agitation.

Laboratory tip: Add Peptide Matrix Water™ to your basket — the recommended stabilising solvent for all reconstituted peptides.

Laboratory protocol — reconstitution, research protocols, storage

Standard protocol, applicable to all research peptides in the Peptology catalogue. For the concentration of the solution (mg/mL) per vial, use the peptide reconstitution calculator.

The 60-second protocol

2. RECONSTITUTION

The solvent is added to the wall of the vial, without agitation, then wait 10–15 minutes.

3. CALCULATION

Calculator → concentration in mg/mL

4. STORAGE

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water

Quick reference

Parameter Value
Form Lyophilised powder
Solvent Peptide Matrix Water
Volume for reconstitution 1–3 mL — 3 mL maximum (capacity of the standard vial)
Storage before reconstitution Room temperature, protected from direct light (refrigeration is not mandatory)
After reconstitution Up to 60 days at +2 °C to +8 °C with Peptide Matrix Water

What you will need

Precision pipette

Calibrated, 100–1000 µL — for accurate measurement of the volume of solvent added to the vial.

70% alcohol pads

Two pads — to disinfect the peptide vial and the solvent vial.

Peptide Matrix Water

Stabilised solvent for lyophilised peptides. Why this one.

Five rules for reconstitution

✅ Correct ❌ Wrong
Disinfect the stoppers and the work surface with 70% alcohol, work with gloves Working without disinfection
The solvent is added slowly down the wall of the vial Direct jet onto the powder — foaming and denaturation
Light swirling between the palms Shaking — foam, on which the peptides denature
Wait 10–15 minutes at room temperature Mechanical mixing or heating
Visual inspection: clear, without particles Using a cloudy or coloured solution

Calculating the concentration

⚠️ Maximum 3 mL of solvent. The standard vial does not allow a larger volume — do not exceed it during reconstitution.

  • Concentration (mg/mL) = mg in the vial ÷ mL of solvent
  • Volume per sample (mL) = amount per sample (mg) ÷ concentration (mg/mL)
  • 1 mg/mL = 1 µg/µL · 1 mL = 1000 µL

👉 For any vial and solvent volume — use the calculator. It calculates the concentration (mg/mL, µg/µL) and the volume per sample (mL, µL).

Storage

BEFORE RECONSTITUTION

Room temperature, protected from direct light, in the original packaging. Refrigeration is not mandatory.

AFTER RECONSTITUTION

+2 °C to +8 °C, up to 60 days with Peptide Matrix Water. Discard the remainder after this period.

FREEZING

Not recommended. Freeze/thaw cycles degrade the structure of the peptide.

Four mistakes to avoid

1. Shaking for “faster dissolution”

Foam = denaturation. Only gentle swirling between the palms.

2. Unsuitable solvent

For the stability of the peptide structure, use only a sterile, purpose-designed solvent — Peptide Matrix Water is recommended.

3. Ignoring slight cloudiness

If it is not completely clear = the sample does not meet the criteria for work.

4. Removing it from the refrigerator “for an hour or two”

Short exposures add up and jeopardise stability.

Research status: All Peptology products are intended exclusively for laboratory use and research.
