FOXO4-DRI (D-Retro-Inverso) is a synthetic peptide of the class of senolytics — compounds studied for the selective removal of senescent (“zombie”) cells from tissues. In the catalogue the name “FOXO4” in practice means FOXO4-DRI, as it is the only commercially relevant peptide of the family.
“DRI” stands for D-Retro-Inverso — a special design in which the peptide consists of D-amino acids in reverse order. This preserves the spatial shape needed for binding to the target, but makes the molecule resistant to proteases and more stable inside the cell than an ordinary L-peptide — so that it reaches its intracellular target and remains active.
Senolytics are among the most discussed fields in ageing biology. With age, senescent cells accumulate in tissues; they stop dividing but remain metabolically active and secrete pro-inflammatory signals (the so-called SASP). FOXO4-DRI is studied as a tool for the selective elimination of these cells without harming healthy ones.
At Peptology it is supplied as a lyophilised research peptide with purity above 99% (see certificates), with a price in Cyprus and pay on delivery. Exclusively for research purposes; it is not a medicine. See also the other peptides for energy and regeneration.
In senescent cells, the FOXO4 protein binds and holds the tumour suppressor p53 in the nucleus, blocking programmed cell death and keeping the cells alive. FOXO4-DRI competitively disrupts the FOXO4–p53 complex, releasing p53 towards the mitochondrial pathway of apoptosis — selectively in senescent cells, without affecting healthy ones.
FOXO4–p53
The target is the FOXO4–p53 interaction, which in senescent cells holds p53 in the nucleus and blocks apoptosis.
SENOLYSIS
The released p53 moves to the mitochondria and triggers selective apoptosis of senescent cells.
DRI STABILITY
The D-retro-inverso structure provides resistance to degradation and a longer intracellular lifetime.
The key idea is selectivity. Healthy cells do not rely on the FOXO4–p53 axis to survive, whereas senescent cells use exactly this interaction to avoid apoptosis. Thus disruption of the complex mainly affects senescent cells — “targeted apoptosis of senescent cells”.
Ordinary peptides are degraded quickly by enzymes. The DRI modification (D-amino acids in reverse order) preserves the three-dimensional orientation required for binding, but makes the peptide resistant to proteolysis. The result is a longer intracellular half-life and a greater chance that the molecule reaches its nuclear target — a critical prerequisite for senolytic action.
Cellular senescence is a state in which the cell stops dividing but does not die. It remains metabolically active and secretes a cocktail of pro-inflammatory molecules — the so-called SASP (Senescence-Associated Secretory Phenotype). With age they accumulate and are linked to chronic inflammation and deterioration of tissue function.
DIVISION ARREST
The cell loses the ability to divide but remains alive and active.
SASP SIGNALS
It secretes pro-inflammatory cytokines that affect neighbouring cells.
ACCUMULATION
Their number rises with age — a subject of study in ageing research.
Senolytics such as FOXO4-DRI are studied precisely to “clear” accumulated senescent cells, so that the effect on tissue function can be studied in preclinical models.
The table compares FOXO4-DRI with other peptides studied in the context of ageing and cellular energy. Each acts through a different mechanism — FOXO4-DRI is the only true senolytic of the group.
| FOXO4-DRI | Epithalon | NAD+ | |
|---|---|---|---|
| Category | Senolytic | Telomeric peptide | Cofactor / energy |
| Mechanism | FOXO4–p53 | Telomerase | Mitochondria |
| Target | Senescent cells | Telomeres / pace | Cellular energy |
| Status | Intermittent | In cycles | Periodic |
The comparison is made by mechanism, for scientific reference; it is not a claim of comparative efficacy, as direct comparative studies are lacking.
The publication by Baar et al. in Cell (2017) — “Targeted apoptosis of senescent cells restores tissue homeostasis” — is foundational for FOXO4-DRI. In aged mice they report improved physical endurance, greater coat density and improved kidney markers after administration.
More recent structural studies (2025) describe in more detail how FOXO4-DRI binds to the transactivation region of p53, clarifying the molecular mechanism of disruption of the FOXO4–p53 complex.
Subsequent research also examines FOXO4-DRI in keloid fibroblasts, chondrocytes, endothelial and brain ageing, broadening the preclinical field.
All available information comes from preclinical (animal and cell) models. There are no clinical data on safety and efficacy in humans — all claims are in a research context.
The directions concern published preclinical studies and are not a claim of results or a recommendation for use.

For reconstitution we recommend Peptide Matrix Water™ — a stabilising solution for reconstituted research peptides; alternatively, bacteriostatic water.
Standard protocol, applicable to all research peptides in the Peptology catalogue. For the concentration of the solution (mg/mL) per vial, use the peptide reconstitution calculator.
1. SOLVENT
2. RECONSTITUTION
The solvent is added to the wall of the vial, without agitation, then wait 10–15 minutes.
3. CALCULATION
Calculator → concentration in mg/mL
4. STORAGE
+2 °C to +8 °C, up to 60 days with Peptide Matrix Water
| Parameter | Value |
|---|---|
| Form | Lyophilised powder |
| Solvent | Peptide Matrix Water |
| Volume for reconstitution | 1–3 mL — 3 mL maximum (capacity of the standard vial) |
| Storage before reconstitution | Room temperature, protected from direct light (refrigeration is not mandatory) |
| After reconstitution | Up to 60 days at +2 °C to +8 °C with Peptide Matrix Water |
Precision pipette
Calibrated, 100–1000 µL — for accurate measurement of the volume of solvent added to the vial.
70% alcohol pads
Two pads — to disinfect the peptide vial and the solvent vial.
Stabilised solvent for lyophilised peptides. Why this one.
| ✅ Correct | ❌ Wrong |
|---|---|
| Disinfect the stoppers and the work surface with 70% alcohol, work with gloves | Working without disinfection |
| The solvent is added slowly down the wall of the vial | Direct jet onto the powder — foaming and denaturation |
| Light swirling between the palms | Shaking — foam, on which the peptides denature |
| Wait 10–15 minutes at room temperature | Mechanical mixing or heating |
| Visual inspection: clear, without particles | Using a cloudy or coloured solution |
⚠️ Maximum 3 mL of solvent. The standard vial does not allow a larger volume — do not exceed it during reconstitution.
Reconstitution per vial — solvent: Peptide Matrix Water. Concentration after addition (1 mg/mL = 1 µg/µL):
| Vial | + 1 mL | + 2 mL | + 3 mL |
|---|---|---|---|
| 10 mg | 10 mg/mL | 5 mg/mL | 3.33 mg/mL |
👉 For any vial and solvent volume — use the calculator. It calculates the concentration (mg/mL, µg/µL) and the volume per sample (mL, µL).
BEFORE RECONSTITUTION
Room temperature, protected from direct light, in the original packaging. Refrigeration is not mandatory.
AFTER RECONSTITUTION
+2 °C to +8 °C, up to 60 days with Peptide Matrix Water. Discard the remainder after this period.
FREEZING
Not recommended. Freeze/thaw cycles degrade the structure of the peptide.
1. Shaking for “faster dissolution”
Foam = denaturation. Only gentle swirling between the palms.
2. Unsuitable solvent
For the stability of the peptide structure, use only a sterile, purpose-designed solvent — Peptide Matrix Water is recommended.
3. Ignoring slight cloudiness
If it is not completely clear = the sample does not meet the criteria for work.
4. Removing it from the refrigerator “for an hour or two”
Short exposures add up and jeopardise stability.
